Nicotinamide, also known as niacinamide, is the amide derivative of nicotinic acid (niacin) and a form of vitamin B3, a water-soluble essential nutrient that serves as a precursor to the coenzymes nicotinamide adenine dinucleotide (NAD⁺) and nicotinamide adenine dinucleotide phosphate (NADP⁺), which are critical for cellular energy production, redox reactions, and metabolic processes. Discovered in the 1930s as a treatment for pellagra, it is chemically 3-pyridinecarboxamide with the molecular formula C₆H₆N₂O and a molecular weight of 122.12 g/mol; it appears as a white crystalline powder that is highly soluble in water (1 g/mL) and has a melting point of 128.8–130 °C.[1][2] Found naturally in foods such as fish, poultry, and yeast, nicotinamide prevents niacin deficiency diseases like pellagra in humans and blacktongue in animals by replenishing NAD⁺ levels.[3][4]
