Methotrexate, formerly known as amethopterin, is a chemotherapy agent and immune-system suppressant. It is used to treat cancer, autoimmune diseases, and ectopic pregnancies. It is used for cancers such as breast cancer, leukemia, lung cancer, lymphoma, gestational trophoblastic disease, and osteosarcoma, as well as autoimmune diseases such as psoriasis, rheumatoid arthritis, and Crohn's disease. It can be given by mouth or by injection.
Methotrexate หรือชื่อเดิม amethopterin เป็นยาเคมีบำบัดและยากดภูมิคุ้มกันของร่างกาย ใช้สำหรับรักษาโรคมะเร็ง โรคภูมิคุ้มกันทำลายตนเอง และการตั้งครรภ์นอกมดลูก ใช้ในโรคมะเร็ง เช่น มะเร็งเต้านม มะเร็งเม็ดเลือดขาว (leukemia) มะเร็งปอด มะเร็งต่อมน้ำเหลือง (lymphoma) โรคถุงน้ำเลื้อยพลาเซนตา (gestational trophoblastic disease) และมะเร็งกระดูก (osteosarcoma) รวมถึงโรคภูมิคุ้มกันทำลายตนเอง เช่น โรคสะเก็ดเงิน (psoriasis) โรคข้ออักเสบรูมาตอยด์ (rheumatoid arthritis) และโรคโครห์น (Crohn's disease) สามารถให้ยาได้โดยการรับประทานหรือการฉีด

Methotrexate is a synthetic folic acid analogue that functions as an antimetabolite by competitively inhibiting dihydrofolate reductase (DHFR), thereby preventing the regeneration of tetrahydrofolate and disrupting the synthesis of purine and pyrimidine nucleotides essential for DNA and RNA production in rapidly dividing cells.[1][2] Developed in 1949 as a structural congener of folic acid to interfere with cellular replication, it was approved by the U.S. Food and Drug Administration in 1953 for the treatment of certain cancers.[2][3]
